Mitochondrial Genetics. Xii. an Oligomycin-Resistant Mutant Localized at a New Mitochondrial Locus in SACCHAROMYCES CEREVISIAE.

چکیده
Three antibiotic-resistance mutations were isolated from strain FL496-2B: two are independent Mendelian genes, one conferring both oligomycin and venturicidin resistance (oli(R) (496)) and the other conferring cycloheximide resistance (cyh(R) (496)). The third is a mitochondrial mutation, O(R) (9), and confers a low level of oligomycin resistance to cells (in vivo) but not to the extracted mitochondrial ATPase (in vitro). This mutation is located on the mitochondrial DNA at a new locus [OLI4] linked to [OLI2] and independent from [OLI1] and [OLI3] and from the other mitochondrial loci. All three mutations (O( R) (9), oli(R) (496), cyh(R) (496 )) were found without any selection, in the same prototrophic haploid strain, which contained unknown resistances to antibiotics. Some physiological, genetical and biochemical properties of the mitochondrial mutation are described.

Genetic analysis of multiple drug cross resistance in Saccharomyces cerevisiae: a nuclear-mitochondrial gene interaction.
A mutant of the yeast Saccharomyces cerevisiae, cross resistant to several antibiotics, was isolated in our laboratory and subjected to genetic analysis. Tetrad analysis of diploids obtained from crosses between the resistant mutant and a sensitive wild-type strain suggest that the multiple resistance to the five agents, oligomycin (OLI), rhodamine 6G (RHG), tetracycline (TCN), chloramphenicol ...
متن کاملCisplatin cytotoxicity is dependent on mitochondrial respiration in Saccharomyces cerevisiae
Objective(s): To understand the role of mitochondrial respiration in cisplatin sensitivity, we have employed wild-type and mitochondrial DNA depleted Rho0 yeast cells. Materials and Methods: Wild type and Rho0 yeast cultured in fermentable and non-fermentable sugar containing media, were studied for their sensitivity against cisplatin by monitoring growth curves, oxygen consumption, pH changes ...
متن کاملIdentification of amino acid substitutions in the dicyclohexylcarbodiimide-binding subunit of the mitochondrial ATPase complex from oligomycin-resistant mutants of Saccharomyces cerevisiae.
1. Oligomycin-resistant mutants of Saccharomyces cerevisiae strain D273-10B/A1 have been assigned to three previously defined mitochondrial genetic loci, olil, oli2 and oli3. Standard crosses of o/iJ-resistant mutants yielded a low percentage of sensitive recombinants indicating that the mutations were in closely linked but separate alleles. 2. The oligomycin-resistant mutants were used to stud...
متن کاملCorrelation between oligomycin-sensitivity and ergosterol in a purified mitochondrial adenosine triphosphatase from Saccharomyces cerevisiae.
use of mutants of Saccharomyces cerevisiae that, while maintaining the aerobic growth characteristics of the parent wild-type, show a resistance to oligomycin (Avner & Griffiths, 1970; Griffiths et al., 1972). This resistance appears to be located in the ATPsynthesizing or -hydrolysing enzyme (ATPaset) of the inner mitochondrial membrane. As a less complex material than entire mitochondrial mem...
متن کاملCharacterization of an Interesting Novel Mutant Strain of Commercial Saccharomyces cerevisiae
The yeast strains that are resistant to high concentration of ethanol have biotechnological benefits and aresuitable models for physiology and molecular genetics research fields. A novel ethanol-tolerant mutant strain,mut1, derived from the commercial Saccharomyces cerevisiae showed higher ethanol production, and alsodemonstrated resistance to ethanol but not to other alcohols...
